Rangers take on Celtic at Ibrox meaning that one of the Glasgow rivals will be absent come May.
The only other potential all-SPL clashes involve Hamilton and Dundee United (if they can overcome Ross County, their Cup final opponents from last year) and Hibernian (if they beat Ayr United) and St Mirren (if they knock out Peterhead in their replay).
Elsewhere, high-flying Hearts could be set for a Cup run. If the Edinburgh club, who sit third in the SPL, get past St Johnstone, they will face First Division opposition in either Falkirk or Partick Thistle.
Motherwell were handed the easiest draw, although it will be some time until they learn their opponents: one of Stenhousemuir, Threave Rovers or Stranraer.
In the remaining fixture, East Stirlingshire or Buckie Thistle were drawn against Queen of the South or Brechin City.
All fifth round ties are scheduled to be played on the weekend of 5th and 6th of February 2011.
